RNA sequencing library preparation

Basic Information

Name: RNA sequencing library preparation
Facility: DRESDEN-concept Genome Center (DcGC)
Partner: Technische Universität Dresden (TUD)
Partner: Max Planck Institute of Molecular Cell Biology and Genetics (MPI-CBG)
Partner: University Hospital Carl Gustav Carus Dresden (UKD)

Description

RNA-Sequencing

  • Poly dT based mRNA isolation from total RNA
  • Barcoded strand- and non-strandspecific RNA-Seq library preparation for single read and paired end reads sequencing (? 3 ?g total RNA)
  • Low input RNA-Seq library preparation (< 3 ?g input) down to 500 ng established and further optimized in respect of maximizing the number of exonic reads, minimizing the number of intergenic reads
  • With FACS sorting (see FACS Facility) combined RNA-Seq workflow enabling preparations of less than 500.000 cells (still in test phase, with 1 Mio cells it works perfect)
