Quality Control of pluripotent stem cells

Basic Information

Name: Quality Control of pluripotent stem cells
Facility: Stem Cell Engineering Facility (SCEF)
Partner: Technische Universität Dresden (TUD)

Description

Characterization of iPSCs:

  • ICC for Pluripotency Markers

Immunocytochemistry (ICC) for pluripotency markers typically expressed in iPSCs: OCT4, SOX2, NANOG and TRA-1-60.

  • Flow Cytometry for Pluripotency Markers

Flow Cytometry for markers of the undifferetiated state typically expressed in iPSCs: OCT4, SOX2, SSEA-4 and TRA-1-60.

  • 96-Gene qRT-PCR Array

Quantitative RT-PCR Array for 96 genes involved in pluripotency, stem cell maintenance, differentiation and house keeping.

  • Trilineage Differentiation

Differentiation of iPSC to derivatives of the 3 germ layers: ectoderm, endoderm and mesoderm.

  • Karyotyping

Karyotype analysis by Giemsa staining of metaphase chromosomes.

  • Mycoplasma Test

PCR test for the most common contaminant in cell cultures - Mycoplasma spp.

  • STR Analysis

Short Tandem Repeat (STR) analysis by multiplex PCR for cell line identification.
